Darolutamide is een nieuwe androgeenreceptor (AR) die is ontworpen om de groei van prostaatkankercellen te remmen door binding aan de AR om de door testosteron geïnduceerde AR-nucleaire translocatie te remmen.
Darolutamide wordt vooral gebruikt bij patiënten met progressieve metastatische castratie-resistente prostaatkanker. Het is effectief bij het remmen van de activiteit van sommige mutante AR's die opkomen tijdens antiandrogene therapieën, waaronder de F876L-mutatieversie die resistent is tegen enzalutamide en ARN-509 (de tweede generatie antiandrogenen). Daarom heeft het potentieel om de weerstandsprobleem te overwinnen die zich voordoet op AR-gerichte therapieën.
